RESUMO

The ability of amphipathic peptides to arrange themselves in aqueous solutions, known as self-assembly, has been found to reduce the effectiveness of these peptides in interacting with cell membranes. Therefore, minimizing their tendency to self-assemble could be a potential strategy for enhancing the pharmacological properties of antimicrobial peptides (AMPs). To explore this idea, this study prepared a series of natural peptides mastoparan C (MPC) with increased net charge and hydrophilicity via alanine-to-lysine substitution and investigated the impact on the biological activity. The preliminary data suggested the influence of both the overall positive charge and the position of a lysine residue on the self-assembly of MPC and its derivatives. Besides, the analogue MPC-A5K,A8K displayed higher anticancer activity and comparable antimicrobial activity with significantly lower hemolysis than MPC. Hence, reducing self-assembly by expanding the cationic area could be a promising approach for developing potent and selective AMPs.

Gallbladder rupture caused by blunt abdominal trauma is an uncommon injury, particularly in children. This condition occurs even less frequently in children with common bile duct cysts. The history is difficult to obtain, the clinical symptoms are indistinct, and their assessment is obscured by a slew of other lesions. Radiography cannot produce clear images. Thus, the diagnosis is frequently delayed and confirmed only during surgery. Case reports of gallbladder injury after abdominal trauma are relatively rare and there are only 50 case reports in literature for 100 years. Herein, we present the case of an 18-month-old girl who had a ruptured gallbladder funnel due to blunt abdominal trauma caused by domestic violence, with an early clinical presentation of septic shock and chylous effusion. The patient underwent surgery after being diagnosed with chylous ascites that had not responded to medical treatment. Based on the findings, single-stage laparoscopic surgery is confirmed to aid in the diagnosis and treatment of gallbladder injury in the presence of a common bile duct cyst.

The present study investigated the phytochemicals and antidiabetic effect of the aqueous extract of the fruit of Punica granatum Linn. in streptozotocin (STZ)-induced diabetic mice. The fruit of P. granatum was extracted with water. The phytochemicals of the water extract were investigated by the liquid chromatography-tandem mass spectrometry (LC-MS/MS) method. The water extract of P. granatum at doses of 150 and 300 mg/kg body weight (bw) was administered to mice for 21 days, and blood glucose level, triglyceride, total cholesterol, low-density lipoprotein (LDL)-cholesterol, high-density lipoprotein-cholesterol, aspartate aminotransferase (AST) and alanine aminotransferase (ALT) enzyme were estimated. Analyzed phytochemicals showed the fruit of P. granatum Linn has a high amount of phenolic and flavonoid compounds, which provide beneficial effect for this plant. The oral administration of the fruit extract of P. granatum at doses of 150 and 300 mg/kg bw for 21 days significantly reduced blood glucose level, triglycerides, serum cholesterol, LDL-cholesterol, AST and ALT enzyme. Our results suggested that the fruit extract of P. granatum has strong antidiabetic effect in STZ-induced diabetic mice. The fruit of this plant might be a potential source of drug for treatment of diabetes.

The reproductive performance of Holstein x Lai Sind crossbred dairy heifers and cows was evaluated at smallholdings in Ho Chi Minh City (HCMC). The data of 232 heifers and 244 cows (261 lactations) from 35 small dairy farms was collected over a 2-year period, from 2013 to 2014. The overall mean for age at first service (AFS), days between first and last service (DFLS), and age at conception (AC) of heifers was 479 (±80), 38 (±80), and 517 (±114) days, respectively. Average number of services per conception (NSC), conception rate (CR), and conception rate at first service (CRFS) was 1.8 (±1.4), 55, and 58%, respectively. The overall mean for the waiting period (WP), DFLS, and days open (DO) of cows was 109 (±52), 133 (±114), and 242 (±129) days, respectively. The mean for NSC, CR, and CRFS was 4.3 (±2.7), 23, and 14%, respectively. A very significant decrease in AFS and AC according to the year of birth, and a significant increase in AFS according to body weight at first insemination (>320 kg) were observed. The CRFS of heifers inseminated in 2013 was significantly higher than in 2014. Monthly mean CR in heifers and cows was negatively correlated with THI. The WP, DFLS, DO, and NSC of cows significantly decreased according to the year of calving. Cows that calved in rainy season had a significantly longer WP than in dry season. The occurrence of postpartum (PP) diseases was accompanied by an increase in WP, DO, and NSC. A decrease in body condition score (BCS) between calving and 60th day (C60) and also between 60th and 120th day of lactation resulted in a significant increase in WP and DO. A decrease in BCS between C60 was also accompanied by a considerable reduction in CRFS of cows. Season and BCS at insemination had a significant effect on NSC and CR of cows. Reproductive performance was satisfactory for heifers, but poor for cows. Losses of BCS during the first months of lactation and at insemination were the major risk factors for this poor performance. Moreover, the season had a more negative effect on fertility in cows than in heifers.
